Adeno-associated viruses (AAV) represent the most commonly used viral vector for the delivery of therapeutic transgenes. However, the gene therapy community is facing major challenges like the high demand for efficient production of high quality AAV vectors in clinical scale quantities.

Robust, quality protein standards and fast, automated highly sensitive analytical instrumentation are needed to address these challenges. A comprehensive monitoring of VP1, VP2 and VP3 protein concentrations and their molar ratios is an effective tool to ensure initial optimization and consistent quality of the final AAV gene therapy products.

Here we introduce PROGEN´s recombinant VP standards used with Simple Western automated capillary electrophoresis in fit for purpose solutions for viral titer and VP capsid protein ratio measurements.

We further show the establishment of the AAV2 VP protein standards by PROGEN as well as the assay performance data that demonstrates linearity and high-sensitivity in an automated workflow optimized for AAV samples.

Dr. Dana Holzinger studied biology at the Ruprecht Karls University in Heidelberg, Germany. Her dissertation at the German Cancer Research Center (DKFZ) and the University Hospital in Heidelberg focused on the identification of human papillomavirus-associated head and neck squamous cell carcinomas. Afterwards, she worked as a post-doctoral researcher leading several molecular epidemiological studies on the identification of biomarkers for different HPV-associated pathologies. In 2019, she joined PROGEN located in Heidelberg, Germany as product manager and took over the role as Head of Product Management in 2020.





Dr. Chris Heger, Director of Applications Science for the Analytical Solutions Division of Bio‑Techne, received his doctoral degree in Pharmacology from Cornell University and completed his post-doctoral training at the National Cancer Institute in Antibody Development, Purification and Technology Evaluation. Chris joined ProteinSimple in 2011, starting as a Field Applications Scientist, and subsequently further building the East Coast support team as Field Manager. During these years, Chris developed a broad but deep understanding of the instrumentation and their application across diverse research areas. In 2016, Chris moved to California to lead the Applications Science group, chartered with creating scientific collateral, fostering collaborations, training, and application development. Chris is an expert in immunoassays, capillary electrophoresis, protein and antibody purification, and chromatography.
